In clinical testing projects, blood samples commonly used for biochemical testing, serum testing, and immunological index testing are generally separated using serum separating gel. This is because the separation gel material contained in the tube has stable performance and can effectively separate serum and plasma samples, making it a popular reagent product on the market. However, in order to improve the quality of serum samples, many manufacturers have strict requirements for the gelation process. Below, we will explain how to perform gelation specifically.

1、 Gluing

Gluing can be divided into machine adding and manual adding:

1. When using a fully automatic glue adding machine, the separating glue is first poured into the machine hopper. After the glue gradually flows flat, cover the hopper and start feeding. If the winter temperature is too low, water baths or electric heating methods can be used to increase the temperature appropriately. The temperature should not be too high and should be controlled around 30-60 degrees Celsius.

2. When manually adding glue, the colloid will be injected into the feeder, with the head of the syringe aligned with the bottom of the test tube, and slowly pushed in to add. After the colloid is sufficient in the test tube, it can be carefully taken out.

2、 Exhaust bubble

After adding the adhesive, place the test tube containing the separation adhesive upright for about 8-12 hours, and then seal and vacuum it. During the manual gluing process, gas will enter and a large number of bubbles will appear during vacuum pumping. It is recommended to use a centrifuge to centrifuge the gas and reduce bubbles. The specific time can be adjusted according to the actual situation. In addition, it is important to first add separation gel, then exhaust bubbles, and then add other types of reagents before vacuuming, so as not to cause interference with other reagents.

3、 Quiet

Whether adding glue or vacuuming and centrifuging, the thixotropy of the separating glue will be temporarily reduced. Therefore, after adding glue and centrifuging, the test tube should be kept upright for more than 1 hour to facilitate the recovery of the thixotropy of the separating glue and to fully release the air in the glue.

4、 Centrifugal condition

Due to the need for centrifugation after completing the collection of blood samples, it is important to emphasize the requirements for centrifugation conditions:

1. Preparation of plasma: After collecting the blood sample, it is necessary to immediately invert and mix it 5-8 times to allow the blood to fully come into contact with the reagent in the tube. Then, centrifuge it with 1800-2200g of centrifugal force for about 8-10 minutes.

2. Preparation of serum: The centrifugation conditions for preparing plasma are similar, but it is important to wait for the blood to completely coagulate before centrifugation, otherwise hemolysis may occur.
