TAPS,the full name N-tris(hydroxymethyl)methyl-3-aminopropanesulfonic acid, CAS29915-38-6, the ingredients in Good's buffer for biological research, need to be stored at room temperature, protected from light and moisture. Trimethylolmethylaminopropanesulfonic acid (TAPS) is a zwitterionic buffer widely used in the fields of biochemistry and molecular biology, with a pH buffer range of 7.7-9.1, soluble in water (25g/50ml).

In order to insure the company staff better understand the TAPS, the TAPS synthesis experiment was conducted under the leadership of the company's R&D manager Liu, using trimethylolaminomethane (Tris) and 1,3-propane sultone (1,3-PS) as raw materials to make it react in alcohol solvent.

 

Specific operation: Weigh equimolar amounts of Tris and 1,3-PS, and take an appropriate amount of alcoholic solvent into a 250ml three-necked flask with condensing reflux. With stirring, heat up and reflux; after a period of reaction, Stop the reaction; at room temperature, after the reaction solution is left to cool to room temperature, suction filtration under reduced pressure, TAPS coarse filter cake washed with an appropriate amount of absolute ethanol 2~3 times, take out the TAPS filter cake, put it in a vacuum drying oven and dry it, and the filtrate filtered under reduced pressure is collected for use.

In the above synthesis process, the reaction time, different alcohol solvents and the recycling of the reaction solvent have a certain effect on the TAPS yield and purity. The experimental results show that the reaction time is controlled at 6 hours, and the yield is the highest, reaching 70.3%. Continue to extend the reaction time, the yield decreases slightly; for the ethanol, n-propanol, isopropanol and n-butanol reaction solvents investigated, n-butanol The yield and purity of TAPS as the reaction solvent are the highest, 70.3% and 94.36%, respectively; in the reaction process, the recycling of the reaction solvent is beneficial to improve the yield and purity of TAPS.

 

How to prepare TAPS solution (about 1-2L)

(1) With 0.1M solution (A):          24.328gTAPS / 1,000ml deionized water

(2) With 0.1M NaOH solution (B):    4gNaOH/ 1,000mldeionized water

(3) pH 4.6    1,000ml (A) + 0ml (B)       (A) :( B) = 5: 0

     pH 7.8    1,000ml (A) + 200ml (B)     (A) :( B) = 5: 1

     pH 8.3     1,000ml (A) + 400ml (B)     (A) :( B) = 5: 2

     pH 8.6     1,000ml (A) + 600ml (B)     (A) :( B) = 5: 3

     pH 9.0     1,000ml (A) + 800ml (B)     (A) :( B) = 5: 4
