1. Catalyst rapid luminescence method
Because under normal circumstances, the chemiluminescence reaction between hydrogen peroxide and luminol luminescence system is very slow, but after adding some catalysts that will not affect the entire detection system, the reaction will become very rapid. Commonly used luminol catalysts include metal complexes: hemoglobin, peroxidase, and transition metal compounds: Fe3+, Fe2+, Mn2+, Cr2+, etc.
2. Indirect inhibitor determination method
In addition to the above-mentioned catalyst acceleration method, some organic substances can be used as inhibitors to inhibit the chemiluminescence of luminol. For example, reducing compounds containing phenolic hydroxyl groups can react with oxidants during the reaction to reduce the chemiluminescence of luminol. The concentration of oxidant is reduced, and the intensity of chemiluminescence is reduced, so as to indirectly measure this kind of organic substances.
3. Liquid chromatography (LC) separation and detection method
Luminol derivatives such as isoluminol (ABEI) are labeled on carboxylic acid and ammonia compounds, and after separation by high performance liquid chromatography (HPLC) or liquid chromatography (LC), they are then combined with Hydrogen peroxide-potassium ferricyanide reaction for chemiluminescence detection. Other separation methods can also be used, such as labeling the newly synthesized chemiluminescence reagent isoluminol isothiocyanate to yeast RNA, separating it by centrifugation and dialysis, and then performing chemiluminescence detection.
