Human 4-1BB(Receptor induced by lymphocyte activation, ILA)
4-1BB is called receptor induced by lymphocyte activation (ILA), also known as CD137, and is also known as a member of the tumor necrosis factor receptor superfamily 9, TNFRSF9. Costimulatory molecule 4-1 BB and its ligands are members of the tumor necrosis factor receptor superfamily. 4-1BB is a type I transmembrane protein. The mature 4-1BB protein contains 256 amino acids and has a relative molecular mass of 30 kD. It is mainly composed of 163aa extracellular domain, 27aa transmembrane domain and 42aa cytoplasmic domain. The ligand of 4-1BB, 4-1BBL, is a type II transmembrane glycoprotein, mainly expressed in professional antigen-presenting cells, such as B cells, macrophages and dendritic cells (DC). The human 4-1BBL gene encodes a protein consisting of 254 amino acids. There are two types, membrane-bound and soluble, which are biologically active.
The interaction between 4-1BB and its ligand 4-1 BBL can provide a second signal independent of CD28 signal for T cell activation, induce T cell proliferation, differentiation and cytokine production, participate in autoimmune diseases and tumor , Viral infection, transplant rejection and inflammation of the immune regulation process. Its regulatory function and regulatory mechanism have now become a hot spot in immunology research.
