Starting from the initial use of phenol, many types of chromogen substrates have now been developed. The items used in clinical biochemical testing include: glucose GLU, triglyceride GT, cholesterol CHOL, uric acid UA, high-density lipoprotein HDL, low density lipoprotein LDL, etc. The detection of creatinine in the renal function series uses TOPS as a chromogenic substrate.

The principle of TOPS for the detection of renal function and creatinine:

The method of detecting creatinine by TOPS belongs to the enzymatic photometric method. The detection principle is: creatinase hydrolyzes creatinine into creatine; creatinase hydrolyzes creatine to form creatine; sarcosine oxidase oxidizes creatine to produce hydrogen peroxide. In the presence of peroxidase, hydrogen peroxide reacts with 4-aminoantipyrine (4-AAP) and TOPS (ESPMT) or HTIB to produce quinone imines, which are colored products. The color intensity can be measured by absorbance, which is directly proportional to the creatinine concentration.

In the experiment, the standard sample tube for collecting the sample can be a vacuum blood collection tube, and the anticoagulant can be heparin or EDTA dipotassium or tripotassium produced by us. The serum or plasma is separated within 1 hour and tested immediately. Store at : 4℃- 8 ℃ stable for 7 days -20 ℃ stable for 3 months , -70 ℃ stable for a long time.

The chromogen substrate TOPS and POD or HRP enzyme preparations cannot be placed in the air for a long time. They need to be prepared for immediate use or refrigerated.
