The chromogen substrate TOOS is a new type of Trinder’s reagent. Its enzymatic reaction with hydrogen peroxide has high sensitivity and specificity. It is usually used for the determination of indicators that can generate hydrogen peroxide in biochemical detection. For example, TOOS is used in the kit to measure the concentration of uric acid in human serum.

Significance of uric acid determination:

Uric acid is the product of purine metabolism in the body. Adenosine deaminase ADA catalyzes the deamination of adenine nucleoside into inosine nucleoside, and then undergoes a series of enzymatic reactions to finally be metabolized into urea. The level of uric acid reflects the health of the body. For example, reduced uric acid excretion or hyperpurine metabolism may cause hyperuricemia and ventilation; too little uric acid production or excessive excretion can cause hypouricemia. Therefore, it is particularly important to maintain the normal level of uric acid in the body and the determination of uric acid level.

 

TOOS enzymatic determination of uric acid:

Uric acid oxidizes with oxygen and water under the catalysis of uricase to generate allantoin and hydrogen peroxide. In the presence of peroxidase, the coupling of the chromogen substrate TOOS and 4-aminoantipyrine is generated in the previous step. The oxidized hydrogen peroxide produces red quinoneimine substances. The darker the color, the more color-developing products. The content can be measured with a spectrophotometer, and the absorbance is directly proportional to the content of uric acid.

 

In order to facilitate the experiment operation and avoid the trouble of repeatedly adding reagents, the unreacted reagent combination can be divided into two groups of reagents. Reagent 1 is mainly biological buffer, peroxidase, and chromogen substrate TOOS; Reagent 2 is mainly uricase , 4-AAP, sodium azide. In the experiment, only need to mix the two in different proportions.

 

It should be noted that when using TOOS to determine uric acid, the amount of reagents and specimens can be increased or decreased proportionally according to different instruments. If the uric acid content in the specimen is too high (greater than 1190uMol/L), normal saline can be used to compare the specimen one to one. Dilute, then measure again, and multiply the result by 2.
