The biological buffer TAPS is a biochemical reagent commonly used in biochemical experiments to maintain the pH of the reaction system, and is widely used in experiments on nucleic acid, DNA and RNA. Its full name is N-tris(hydroxymethyl)methyl-3-aminopropanesulfonic acid CAS: 29915-38-6, and the buffer range is 7.7-9.1.

TAPS molecule contains 3 hydroxyl groups, which has good hydrophilicity and is easily soluble in water. The soluble TAPS can reach 50g per 100 grams of water, so TAPS can be directly dissolved in deionized water to prepare a solution. TAPS can be regarded as a tris hydroxymethylaminomethane Tris with a propanesulfonic acid group attached to the amino group, which can be synthesized by Tris and 1,3-propane sultone in alcohol solution.

When TAPS is formulated into a buffer solution, sodium hydroxide lye is usually used to adjust the pH. First, weigh the TAPS powder and solid sodium hydroxide (the relative molecular mass of TAPS is 243.28, and that of sodium hydroxide is 40), and then they are prepared into solutions and labeled A and B.

Biological buffer TAPS reagent powder

Adjust the mixing ratio of the two solutions AB according to the required pH value with reference to the following ratio:

pH 4.6 1,000ml(A)+0ml(B) (A):(B)=5:0

pH 7.8 1,000ml(A)+200ml(B) (A):(B)=5:1

pH 8.3 1,000ml(A)+400ml(B) (A):(B)=5:2

pH 8.6 1,000ml(A)+600ml(B) (A):(B)=5:3

PH 9.0 1,000ml(A)+800ml(B) (A):(B)=5:4

If the reaction system needs to exclude sodium ions, potassium hydroxide is used instead of sodium hydroxide, and the temperature is controlled at room temperature 20 degrees when preparing the solution. If you need to prepare a more accurate pH buffer system, you can first prepare it as described above, then measure it with a pH meter, and then slowly add liquid A or liquid B to adjust.

APS is also commonly used in conjunction with other reagents, such as 10X Tris-TAPS-EDTA buffer, which contains 500 mM Tris; 500 mM TAPS; 10 mMEDTA. Tris is alkaline, and when used with TAPS, no other acid is needed.

In addition to DNA screening and RNA sample experiments, TAPS can also be used as a protective agent for oxygenated hemoglobin during freeze-drying, which can effectively protect oxygenated hemoglobin from oxidation to methemoglobin and ensure its oxygen-carrying function. Capillary zone electrophoresis can also use TAPS as background electrolyte and running buffer for protein microanalysis.

The TAPS produced by Desheng is synthesized with Tris and 1,3-PS. Because the company produces more Tris, it has advantages in cost and product quality, whether it is for domestic or foreign customers. A better choice.
