You may not know that when doctors diagnose kidney function in patients based on indicators such as serum creatinine and urea nitrogen, little is known about a colorimetric reagent called DAOS (CAS number: 82692-97-5) playing the role of a "silent guardian". This core material commonly used in uric acid and kidney function testing kits provides a solid foundation for disease monitoring due to its excellent water solubility, sensitivity, and stability. (1) Packaging method
When using DAOS, it is recommended to follow the principle of "small quantity multiple times" for packaging. For milligram level dosage requirements, pre pack the reagents into well sealed bottles and only take a single dose at a time. This operation not only reduces the contact between the reagent and moisture in the air, but also avoids moisture absorption and deterioration caused by repeated bottle opening, ensuring the stability of the test results.
(2) Low temperature storage
DAOS needs to be stored at a low temperature of 0-5 ℃, but it should be placed at room temperature half an hour before use. This process is crucial: if reagents stored at low temperatures are directly used for detection, the enzyme activity may be affected by sudden temperature changes, resulting in insufficient color reaction. After restoring room temperature, the activity of the reagent molecules reaches a better state, which can more accurately capture the target substance in the sample.
(3) Store in the dark
Unused DAOS should be stored in a light shielded container. Light exposure not only accelerates reagent decomposition, but also may cause structural changes in the chromophore, directly affecting detection sensitivity. It is recommended that the laboratory establish a standardized reagent ledger to record the opening time, amount taken, and operators of each batch of reagents, in order to achieve product traceability.
(4) Cold chain transportation
Special attention should be paid to the transportation of DAOS. The general method is to tightly wrap the reagent bottle with foam glue in the inner layer, and place sufficient ice bags in the middle layer to prevent the product from being wet by the water of ice cubes. Winter transportation can reduce the number of ice packs appropriately, but absorbent cotton should be used to isolate condensed water and ensure that the reagents are always in a dry and low-temperature environment.
Detection Principle
Under conditions containing hydrogen peroxide and peroxidase, the novel Trinder's reagent can undergo oxidative coupling reaction with 4-aminoantipyrine (4-AA) or 3-methylbenzothiazole sulfone hydrazone (MBTH) to produce highly stable purple or blue dyes. The substrate is catalyzed by a specific oxidase to produce hydrogen peroxide, and the concentration of this hydrogen peroxide directly reflects the concentration of the original substrate. Therefore, the content of the substrate can be quantitatively analyzed by detecting this color reaction. This method utilizes the color change of oxidative coupling reaction to achieve measurement of the target substance.
