Acridinium ester is a chemiluminescent marker commonly used in fields such as immunoassays and molecular diagnostics. Due to its photosensitivity and instability, special attention should be paid to the following when weighing:
The packaging methods used vary depending on the amount of packaging
When there is a large amount of packaging (such as milligrams), directly use the original/packaging bottle for weighing:
1.1. Place the sorting bottle (such as a brown glass bottle or a light shielded EP tube) on an analytical balance, remove the skin and reset to zero.
1.2. Slowly add acridine ester powder with a clean spoon or a small spoon to avoid dust.
Once the target weight is reached, immediately seal the bottle cap tightly and store it in a dark place.
Advantages: Avoiding losses during the transfer process.
When the packaging volume is small (such as microgram level), use weighing paper to assist in transfer:
2.1 Fold the weighing paper (smooth side up) into a V-shaped groove and place it on the balance to remove the skin.
2.2. Be careful when adding acridine ester powder and record the weight.
2.3 Tilt the weighing paper and gently sweep the powder into the dispensing bottle using a dry fine brush or a small spoon (the bottle mouth can be aligned with the paper slot in advance to reduce flying).
Key point: Avoid breathing and airflow disturbances that may cause powder to scatter.
Other precautions
1. Avoid light operation
Acridine esters are sensitive to light, especially ultraviolet and strong light. When weighing, it should be done in a dark environment (such as under yellow or red lights) or in a light shielded container to avoid direct exposure to natural light or laboratory white light.
2. Control temperature and humidity
Acridine esters are prone to moisture absorption and decomposition, and should be operated in low humidity environments such as dryers or glove boxes. If the laboratory humidity is high, the reagents can be equilibrated to room temperature in advance to avoid the influence of condensation water.
3. Use inert solvents and containers
Before weighing, ensure that the containers used (such as weighing dishes, EP tubes) are dry and clean, avoiding residual organic solvents or moisture interference. It is recommended to store in brown glass bottles or dark plastic containers.
4. Quick weighing to reduce exposure time
Acridine esters are easily degraded in air, so they should be weighed quickly and immediately sealed for storage to reduce contact time with air.
5. Accurate weighing tools
Use a high-precision analytical balance (recommended accuracy ≥ 0.01 mg), as acridine esters are usually used in very small amounts (in the microgram range), errors may significantly affect experimental results.
Following the above steps can maintain the stability of acridine ester and ensure the accuracy of experimental results. For specific operation suggestions, please refer to the product manual or literature methods.
In chemiluminescence detection, the stability and labeling efficiency of acridine esters directly affect the experimental results. Hubei Xindesheng Materials focuses on the research and production of acridine esters and other chemiluminescence reagents. The products have strong stability, high sensitivity, and small batch differences. They can provide acridine esters modified with different active groups (such as NHS esters, carboxyl groups, etc.) to meet the labeling needs of biological molecules such as antibodies and nucleic acids.
