In the biomedical field, catalase (CAT) is a crucial enzyme class, and its activity detection has profound significance for the diagnosis, treatment, and drug development of diseases. Step 2, oxygen donor (TOOS) was introduced as a colorimetric reagent. Under the action of CAT, the remaining H2O2 undergoes an oxidation reaction with TOOS, generating a blue quinone imine substance. By measuring the absorbance of the substance at a wavelength of 555nm, we can accurately calculate the activity level of CAT.

Firstly, in the selection of reaction conditions, we optimized key factors such as pH value, temperature, and substrate concentration, and determined pH 7.5, 30 ℃, and 16.2 mmol · L-1 H2O2 concentration as the reaction conditions. This combination not only ensures the smooth progress of the reaction, but also greatly improves the accuracy and stability of the detection.
Secondly, TOOS serves as a chromogenic substrate, making the reaction process faster and more sensitive. In just a few minutes, the detection of CAT activity can be completed, greatly reducing the detection time and improving work efficiency. At the same time, the color rendering effect of TOOS is clear and concise, making the result judgment more intuitive and accurate.
In addition, this method also performs well in sensitivity and stability. Through meticulous experimental design and optimization, we have successfully raised the detection sensitivity to a higher level, enabling accurate capture even in the presence of trace amounts of CAT.
