Parenzyme (EC 184.108.40.206) is a kind of protease. It is a serine proteolytic enzyme extracted from the pancreas of cattle, sheep and pigs. In vertebrates, it acts as a digestive enzyme. After trypsinogen, the precursor of trypsinase, is synthesized, secreted as a component of pancreatic juice, and decomposed into activated trypsinase restricted by enterokinase or trypsinase. It is an endonuclease that cuts off the carboxyl side of lysine and arginine residues in the polypeptide chain. It not only plays the role of digestive enzymes, but also restricts the decomposition of precursors of chymotrypsinogen, carboxypeptidase, phospholipase and other enzymes, and plays an active role. It is the most specific protease and becomes an indispensable tool in determining the amino acid arrangement of proteins.
Trypsinogen is composed of more than 220 amino acids with 6 pairs of disulfide bonds and its molecular weight is about 24 kD. The serine residues in the active site are indispensable serine proteases. Under the active self-catalysis of enterokinase, the peptide bond between N-terminal lysine and isoleucine residue of proenzyme was hydrolyzed, and the valerian-tian-tian-lys6 peptide was released to produce active trypsin. Trypsin can be used for cell digestion. In addition, trypsinase is one of the instrumental enzymes necessary for insulin production.
