Eye drops are one of the most commonly used drug formulations for ophthalmological diseases. For many eye diseases, eye drops have a direct and rapid therapeutic effect. Now let’s introduce two chemical reagents that are widely used in eye drops, namely TRIS and Carbomer. Let’s take a look at the application of these two substances in eye drops.

Application of TRIS in eye drops

Due to the special anatomical and physiological characteristics of the eyes, corresponding requirements are put forward for ophthalmic preparations: the pH of normal tears is 7.2 to 7.4, and the range of pH tolerated by normal eyes is 5.0 to 9.0. The pH value of the eye drops will directly affect the eye irritation and the curative effect of the drug. When preparing eye drops, a buffer should be added to adjust the pH of the solution so that it is roughly similar to the tear fluid to reduce the impact on the eyes. Stimulus. At the same time, adjusting the pH value also needs to take into account multiple factors such as drug solubility and stability. Tris (Tris) is a commonly used pH regulator in eye drops.

As an organic amine, Tris can form a water-soluble double salt with insoluble drugs with acidic groups. The most common eye drops containing Tris are ketorolac tromethamine eye drops. Ketorolac is a non-steroidal anti-inflammatory drug for injection. Ketorolac tromethamine is soluble in water and stable in aqueous solution, with antipyretic analgesic and anti-inflammatory effects. It reversibly inhibits cyclooxygenase and deoxygenase, thereby inhibiting the biosynthesis of prostaglandins, thereby reducing the response of painful nerves to endogenous inflammatory factors, inhibiting peripheral nerve sensitization, and achieving the purpose of advanced analgesia. Ketorolac tromethamine eye drops are often used for inflammation and pain after eye surgery.

Application of carbomer in eye drops

Carbomer can be used as an ophthalmic gel. The common eye drops currently in clinical use cause poor drug absorption due to the short retention time of the drug in the eye. But Carbomer is different. It not only does not have the above shortcomings, but also clinically it can relieve endocrine disorders and dry eye symptoms caused by insufficient tear production, reduce inflammation and treat presbyopia. The bromhexine hydrochloride eye drops that are clinically used to treat human dry eye disease have poor efficacy due to their short contact time with the cornea. However, the use of a gel made with carbomer has a significant effect of promoting tearing. The maximum effect is produced within 1 hour after use. It should be noted that carbomer is used as a pharmaceutical excipient and does not have other drug effects. It is only used as a gel, thickener, dispersant or suspending agent as a carrier medium for drugs.
